Chemical Composition of Ligusticum Wallichii(Chuan Xiong)

Contains volatile oil, ferulic acid, as well as 4-hydroxy-3-butylphthalide, senkyunolide, Ligustilide, tetramethylpyrazine, chuanxiongol, sedanic acid, etc.

Pharmacological Action of Ligusticum Wallichii(Chuan Xiong)
1. Chuanxiong has a significant sedative effect on the central nervous system. When the volatile oil of Chuanxiong is in small amounts, it has an inhibitory effect on the activity of the animal brain, while it has an excitatory effect on the respiratory center, vascular motor center, and spinal reflex center of the medulla oblongata. Chuanxiong decoction can inhibit the spontaneous activity of rats and mice, prolong the sleep time caused by pentobarbital sodium, and counteract the excitatory effect of caffeine (20mg/kg) when administered separately. But it cannot resist seizures caused by pentylenetetrazol in rats. Administering Chuanxiong decoction 25-50g/kg orally can inhibit the spontaneous activity of rats, and has a more significant sedation effect on larger rats; It can also prolong the sleep time of pentobarbital, but it cannot antagonize the excitement of caffeine, nor prevent the convulsive or lethal effects of pentamethylenetetrazolium and cocaine. The volatile oil of Japanese produced Ligusticum chuanxiong has an inhibitory effect on the activity of the animal brain, while it has an excitatory effect on the vascular motor center, respiratory center, and spinal cord reflex in the medulla oblongata. When the dosage is increased, it becomes inhibited.

2. Effects on the cardiovascular system: Chuanxiong decoction increases the contraction amplitude and slightly slows down the heart rate of isolated toads and frog hearts at concentrations ranging from 1 to 5 to 10 to 4.

3. Effect on smooth muscle: The 10% aqueous solution of Chuanxiong extract can stimulate the isolated uterus of pregnant rabbits in small amounts, causing an increase in tension and contraction, ultimately leading to contracture; A large amount can cause uterine paralysis and stop contractions. Ligusticum chuanxiong alkaloids, ferulic acid, and ligustilide all have spasmolytic effects, while ligustilide is the main component of spasmolysis. Chuanxiong indole ig300mg/kg has a significant analgesic effect in mice, with P<0.001 compared to the control group.

4. In vitro experiments such as antibacterial effects. Chuanxiong has inhibitory effects on Escherichia coli, dysentery (Songnei’s disease), Proteus, Pseudomonas aeruginosa, typhoid fever, paratyphoid bacteria, and Vibrio cholerae. Chuanxiong water extract (1:3) also has inhibitory effects on certain pathogenic skin fungi in vitro.

5. Anti radiation effect: Chuanxiong decoction has a certain therapeutic effect on experimental treatment of animal radiation diseases. The water-soluble crude preparation of Chuanxiong has a protective effect on radiation exposure and nitrogen mustard damage in rats, mice, and dogs. The anti radiation effect of Chuanxiong on rats is better than that of mice, and the effect of ip administration is better than that of im administration. The effect of im administration is better than that of ig administration.

6. Other effects: Ligustrazine can increase renal blood flow and diuresis in anesthetized rabbits. Ligustrazine can inhibit DNA synthesis, indicating that it can inhibit protein and antibody production. Chuanxiong has certain anti vitamin E deficiency effects, which can protect chicks from nutritional encephalopathy caused by vitamin E deficiency. Sodium ferulate can reduce the lipid peroxidation reaction caused by H2O2 and O2, and has anti OH and malondialdehyde (MDA) hemolysis effects. Sodium ferulate can significantly reduce complement hemolysis and inhibit the binding of complement 3b (C36) to the red blood cell membrane. Ligustrazine has an inhibitory effect on the occurrence of pulmonary fibrosis in mice prepared by intratracheal administration of Pingyangmycin.
